Different levels of hsp70 and hsc70 mRNA expression in Iberian fish exposed to distinct river conditions Genet. Mol. Biol.
Jesus,Tiago F.; Inácio,Ângela; Coelho,Maria M..
Comprehension of the mechanisms by which ectotherms, such as fish, respond to thermal stress is paramount for understanding the threats that environmental changes may pose to wild populations. Heat shock proteins are molecular chaperones with an important role in several stress conditions such as high temperatures. In the Iberian Peninsula, particularly in Portugal, freshwater fish of the genus Squalius are subject to daily and seasonal temperature variations. To examine the extent to which different thermal regimes influence the expression patterns of hsp70 and hsc70 transcripts we exposed two species of Squalius (S. torgalensis and S. carolitertii) to different temperatures (20, 25, 30 and 35 ºC). At 35 ºC, there was a significant increase in the...

Effects of hypoxia storage on gene transcript accumulation during tomato fruit ripening Braz. J. Plant Physiol.
Pegoraro,Camila; Santos,Railson Schreinert dos; Krüger,Mariana Madruga; Tiecher,Aline; Maia,Luciano Carlos da; Rombaldi,Cesar Valmor; Oliveira,Antonio Costa de.
Tomato (Solanum lycopersicum L.) is a climacteric fruit, i.e., during ripening an increase in ethylene synthesis and high rate of respiration are observed. Low oxygen levels might inhibit or block ethylene biosynthesis and therefore retard the ripening process. Despite commercial applications of low oxygen treatments, the precise mode of action of low oxygen in fruit tissues and ripening is not well understood. In order to delineate the molecular responses to low oxygen stress in fruits, hypoxia-responsive tomato genes encoding heat shock factors, heat shock proteins, and enzymes involved in fermentation and ethylene synthesis pathways were analyzed. In this study, tomato fruit stored under hypoxia conditions showed that HSP17.7 and HSP21 genes were highly...

Identification and in silico analysis of the Citrus HSP70 molecular chaperone gene family Genet. Mol. Biol.
Fietto,Luciano G.; Costa,Maximiller D.L.; Cruz,Cosme D.; Souza,Alessandra A.; Machado,Marcos A.; Fontes,Elizabeth P.B..
The completion of the genome sequencing of the Arabidopsis thaliana model system provided a powerful molecular tool for comparative analysis of gene families present in the genome of economically relevant plant species. In this investigation, we used the sequences of the Arabidopsis Hsp70 gene family to identify and annotate the Citrus Hsp70 genes represented in the CitEST database. Based on sequence comparison analysis, we identified 18 clusters that were further divided into 5 subgroups encoding four mitochondrial mtHsp70s, three plastid csHsp70s, one ER luminal Hsp70 BiP, two HSP110/SSE-related proteins and eight cytosolic Hsp/Hsc70s. We also analyzed the expression profile by digital Northern of each Hsp70 transcript in different organs and in response...

Identification and in silico expression pattern analysis of Eucalyptus expressed sequencing tags (ESTs) encoding molecular chaperones Genet. Mol. Biol.
Cagliari,Thiago C.; Tiroli,Ana O.; Borges,Júlio C.; Ramos,Carlos H.I..
Expressed Sequence Tags (ESTs) sequencing provides reliable and useful information concerning gene expression patterns in the genomic context. Our group used bioinformatics to identify and annotate 5'EST-contigs belonging to the molecular chaperones within the Eucalyptus Genome Sequencing Project Consortium (FORESTs) database. We found that 1,959 5'EST-contigs, or approximately 1.6% of the total 5'EST-contigs, encoded chaperones, emphasizing their biological importance. About 55% of the chaperones that we found were Hsp70 chaperones and its co-chaperones, 18% were Hsp90 chaperones, 15% were Hsp60 and its co-chaperone, 8% were Hsp100 chaperones, and 4% were Small Hsps. We also investigated the digital expression profile of the chaperone genes to gain...

Small heat shock proteins genes are differentially expressed in distinct varieties of common bean Braz. J. Plant Physiol.
Simões-Araújo,Jean Luiz; Rumjanek,Norma Gouvêa; Margis-Pinheiro,Márcia.
Plants respond to temperature stress by synthesizing a set of heat shock proteins (HSPs), which may be responsible for the acquisition of thermotolerance. In this study, the induction of small HSPs (sHSPs) in eight common bean varieties was evaluated by Northern blot analysis using the W HSP 16.9 cDNA as heterologous probe. Cowpea was used, as a positive control since this plant, as opposed to common bean, is known to grow well under high temperature regimes such as that found in the Brazilian semi-arid region. After the growth period, the plants were submitted to two h of heat shock at 40 ºC. All varieties tested were able to induce sHSP mRNAs that hybridized with W HSP 16.9 probe. However, significant kinetic differences were found when comparing...

Thermal biology of the deep-sea vent annelied Paralvinella grasslei: in vivo studies ArchiMer
Cottin, D; Ravaux, J; Leger, N; Halary, S; Toullec, J; Sarradin, Pierre-marie; Gaill, F; Shillito, B.
The annelid Paralvinella grasslei is a deep-sea vent endemic species that colonizes the wall of active chimneys. We report here the first data on its thermal biology based on in vivo experiments in pressurized aquaria. Our results demonstrate that P. grasslei survives a 30 min exposure at 30 degrees C, and suggest that the upper thermal limit of this species is slightly above this temperature. The first signs of stress were noticed at 30 C, such as a significant increase in the animal's activity and the expression of HSP70 stress proteins. A preliminary investigation of the kinetics of stress protein expression surprisingly showed high levels of HSP70 proteins as late as 3.5 h after the heat shock. Finally, we provide here the first sequences for vent...
